This antibody binds the extracellular domain of human CD47, which is broadly distributed on normal adult tissues and ovarian tumors, and is especially abundant in certain epithelia and the brain. CD47 (IAP, integrin-associated protein) acts as an adhesion receptor for THBS1 on platelets and modulates integrins. It plays key roles in memory formation and synaptic plasticity in the hippocampus. It also serves as a receptor for SIRPA, preventing maturation of immature dendritic cells and inhibiting cytokine production by mature dendritic cells.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins and regulates their functions. Engagement of CD47 by soluble ligands or counter receptors modulates signaling pathways, including activation of heterotrimeric G proteins. Functionally, CD47 contributes to increased intracellular calcium upon cell adhesion to the extracellular matrix. It acts as a receptor for the C-terminal domain of thrombospondin and may play roles in membrane transport and signal transduction. CD47 is broadly distributed across tissues, reduced on Rh erythrocytes, and serves as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages that prevents phagocytosis of CD47-positive cells. The CD47-CD172a interaction also influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanical response modulation, and neuronal development, being abundant in synapse-rich regions of the brain and retina.
