This antibody binds the extracellular domain of human CD47, which is broadly distributed on normal adult tissues and ovarian tumors, and is especially abundant in some epithelia and the brain.
IAP (integrin-associated protein) functions as an adhesion receptor for THBS1 on platelets and modulates integrins. It plays an important role in memory formation and synaptic plasticity in the hippocampus.
CD47 also acts as a receptor for SIRPA; binding to SIRPA prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins and regulates their functions.
Engagement of CD47 by soluble ligands or counter receptors modulates signaling pathways, including activation of heterotrimeric G proteins.
Functionally, CD47 is involved in increasing intracellular calcium concentration upon cell adhesion to the extracellular matrix.
It also serves as a receptor for the C-terminal cell-binding domain of thrombospondin and may play roles in membrane transport and signal transduction.
CD47 expression is broad across tissues but reduced on Rh erythrocytes.
It acts as a ligand for CD172a (SIRPα), an immune inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells.
The CD47-CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and promotes neuronal development, particularly in synapse-rich regions of the brain and retina.
