Product Specific Information

The 104D2 monoclonal antibody reacts with human CD117 (c-Kit), also known as Steel factor receptor or stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells and mast cells, where it mediates proliferation and differentiation. CD117 is also expressed by melanocytes and contributes to their activation and signaling.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells
  • Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

Target Information

Gene: KIT (c-KIT)
Description: Proto-oncogene encoding a type 3 transmembrane receptor for MGF (mast cell growth factor / stem cell factor). Regulates growth and activation of hematopoietic and non-hematopoietic cells.
Clinical Relevance: Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, AML, and piebaldism. The D816V mutation is a key therapeutic target in neoplastic mast cell disorders.
Expression: Hematopoietic stem cells, germ cells, mast cells, and gastrointestinal Cajal cells.
Function: Upon SCF binding, c-Kit dimerizes and autophosphorylates, activating downstream pathways such as Ras/ERK, PI3K, PLC-γ, and JAK/STAT. Essential for hematopoiesis, stem cell maintenance, and gametogenesis.
