Product Specific Information
The TM-b1 monoclonal antibody reacts with mouse CD122, the 90–110 kDa interleukin-2 receptor beta chain (IL-2Rβ). CD122 is expressed by NK cells and some T cells in the periphery, and is upregulated upon activation. CD122 associates with CD25 and CD132 (common gamma chain) to form the high-affinity IL-2 receptor. TM-b1 inhibits high-affinity IL-2 binding.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry on mouse splenocytes at ≤1 µg/test (100 µL final volume). Cell count may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.
Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended detection using a 450/50 bandpass filter or equivalent.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD122 (Interleukin-2 receptor beta chain) is involved in T cell-mediated immune responses, primarily expressed in the hematopoietic system. It exists in three forms:
- Low affinity: α subunit monomer (no signal transduction)
- Intermediate affinity: α/β heterodimer
- High affinity: α/β/γ heterotrimer
Intermediate and high-affinity forms mediate receptor endocytosis and mitogenic signaling. The CD122 gene encodes the β subunit, a type I membrane protein.
Associated diseases include oligoarticular juvenile idiopathic arthritis and rheumatoid factor-negative juvenile idiopathic arthritis.
