Product Specific Information

The A7R34 monoclonal antibody reacts with mouse CD127, the high-affinity alpha subunit of the IL-7 receptor. CD127 is expressed by immature B cells in the bone marrow, double-negative (CD4⁻CD8⁻), single-positive (CD4⁺ and CD8⁺), but not double-positive thymocytes. Mature peripheral T cells express CD127 at low levels.

A7R34 inhibits IL-7 binding to its receptor and is used in vivo and in vitro to study IL-7 roles in T and B cell development and activation. It also blocks binding of SB/199, another CD127-recognizing antibody.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes, ≤1 µg/test (100 µL final volume).

Super Bright 436 is excited by a violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

CD127 (Interleukin-7 receptor alpha chain, IL7Rα) regulates lymphopoiesis. The active IL7R receptor is an alpha/gamma chain heterodimer.

• The gamma(c) chain activates signal transduction.
• The alpha chain determines specific signaling events.

CD127 promotes proliferation of precursor B cells, thymocytes, and mature CD4⁺/CD8⁺ T cells.
Dysfunction is associated with severe combined immunodeficiency (SCID) and T⁻/B⁻/NK⁺ SCID variants.
