Product Specific Information

The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor. This 60–70 kDa transmembrane protein binds to IL-3 with low affinity by itself and, when associated with CD131 (common beta chain), binds IL-3 with high affinity.
CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.

This antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (final volume 100 µL). Cell number can range from 10⁵ to 10⁸ cells/test.

Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

IL-3Ra (Interleukin-3 receptor subunit alpha) is a member of the type I cytokine receptor family. It forms a heterodimer with the common beta subunit (CDw131) for signal transduction of Interleukin-3.
IL-3Ra is overexpressed on acute myeloid leukemia (AML) blasts compared with normal hematopoietic cells, making it a potential target for novel therapeutic agents.
