Product Specific Information

The ACK2 monoclonal antibody reacts with mouse CD117 (c-Kit receptor), also known as Steel factor receptor or stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells, mast cells, and plays key roles in proliferation and differentiation.

• Reported application: Flow cytometric analysis
• Tested application: Flow cytometry on mouse bone marrow cells (≤0.25 µg/test)
• Recommended careful titration for optimal performance
• Super Bright 436: Excitation at 405 nm (violet laser), emission at 436 nm
• Recommended filter: 450/50 bandpass or equivalent
• For multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions

Target Information

KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for MGF (mast cell growth factor, stem cell factor). It regulates growth and activation of hematopoietic and non-hematopoietic cells.

• Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
• Activation of c-Kit triggers autophosphorylation of tyrosine residues, enabling docking of Grb2 and activation of the Ras/ERK pathway.
• SCF/c-Kit signaling activates PI3K, PLC-γ, and JAK/STAT pathways, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
