Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational changes enabling dimerization and DNA binding to estrogen response elements (ERE). This binding modulates gene transcription through coactivator or corepressor recruitment.
There are two forms of estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and essential for sexual development, reproductive function, and bone health. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
