Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding allows ER to regulate gene transcription through coactivator or corepressor proteins.
There are two main forms of the estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four known estrogen receptor-alpha isoforms (Isoform 1: 66 kDa, Isoform 2: 53 kDa, Isoform 3: 47 kDa, Isoform 4: 35 kDa). Estrogen receptors are widely expressed in various tissues, crucial for sexual development and reproductive function, and also play roles in bone and other tissues. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
