Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a novel protein localized with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during tight junction maturation rather than serving as a structural component. Metadherin is overexpressed in breast cancer tissues and xenografts, with much lower expression in normal breast tissue. It binds to lung vasculature—a common metastasis site—through a C-terminal extracellular domain segment. Blocking this domain or inhibiting Metadherin via siRNA has been reported to reduce breast cancer metastasis.
