Product Specific Information
The SK1 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimer or CD8α/α homodimer. CD8a is present on thymocytes, a subset of mature T cells, and NK cells. CD8 binds MHC class I and interacts with protein tyrosine kinase p56lck, contributing to T-cell development and activation.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.2 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample before combining antibodies with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a key role in T-cell signal transduction, differentiation, and activation. CD8 is expressed on T cells as a disulfide-linked heterodimer (CD8α/β) functioning as a co-receptor for MHC class I molecules, while CD4 serves for MHC-II. CD8 also exists as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Upon MHC-I/peptide complex binding, CD8 recruits Lck kinase, triggering lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
