Product Specific Information

The SK1 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is present on most thymocytes and subsets of mature T cells and NK cells. It binds MHC class I and, through association with tyrosine kinase p56lck, plays a role in T-cell development and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.2 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor for MHC class I molecules, aiding TCR-mediated signal transduction and T-cell activation. CD8 is expressed mainly on T cells as a disulfide-linked heterodimer (CD8αβ) and also as a homodimer (CD8αα) on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation. CTLs play a key role in pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
