Product Specific Information

The SK1 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimers or CD8α/α homodimers. CD8a is found on thymocytes, mature T cells, and NK cells, binding to MHC class I and associating with p56lck for T-cell activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.125 µg) per test.

A test is defined as staining a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor with TCR for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8α is essential for MHC-I binding and CTL activation, playing a critical role in pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
