Product Specific Information

The SK1 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimers or CD8α/α homodimers. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.06 µg) per test, defined as staining a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  Add 5 µL per sample to antibody cocktails before combining with cells.
  For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD8 is a type I transmembrane glycoprotein functioning as a co-receptor with TCR for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8α binds MHC-I and recruits Lck kinase, leading to CTL activation and pathogen/tumor clearance. Differentiation into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
