Product Specific Information

The SK1 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with p56lck, playing roles in T-cell development and activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL per test, 100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample to antibody cocktails or single-color controls.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin receptor family, acting as a co-receptor for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8 engagement triggers Lck recruitment, leading to CTL activation and pathogen/tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
