Target Information

Phosphorylation of receptors by protein kinases is a process that can be reversed by a group of enzymes called protein phosphatases. Coordinated control of kinases and phosphatases provides the cell with the capacity to rapidly switch between phosphorylated and dephosphorylated protein states in response to environmental stimuli. Activation of critical enzymes by kinase phosphorylation alone is not sufficient; phosphatase dephosphorylation provides essential regulatory balance. Dysregulation of these processes can lead to uncontrolled cell growth associated with cancers and developmental disorders.

Protein phosphatases may be regulated by expression levels, subcellular localization, phosphorylation of catalytic or regulatory subunits, and endogenous inhibitors. Because most phosphatases are not highly substrate-specific, alterations in their activity can broadly affect cellular signaling networks and physiological responses.
