Product Specific Information

The eBioH4B4 monoclonal antibody reacts with human CD107b (LAMP-2), a highly glycosylated type I transmembrane protein (~105 kDa) localized in lysosomal/endosomal membranes. It is also expressed on the surface of degranulating T cells, activated platelets, and certain cancer cells. Mutations in CD107b cause Danon disease, a lysosomal glycogen storage disorder.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis
  • Surface staining in flow-based degranulation assay

Applications Tested:

  • Pre-titrated and validated for intracellular staining and flow cytometry using Jurkat cells
  • Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Target Information

LAMP2 (CD107b) is a lysosome-associated membrane glycoprotein involved in lysosomal biogenesis, fusion with phagosomes, and chaperone-mediated autophagy. It is a ~46 kDa precursor that becomes 100–130 kDa after glycosylation. LAMP2 has three isoforms (LAMP-2A, -2B, -2C) and functions with LAMP1 in lysosomal membrane integrity. Deficiency leads to autophagosome accumulation and Danon disease (cardiomyopathy, myopathy).
