Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43, also known as leukosialin, galactoglycoprotein, and sialophorin. CD43 is a sialomucin with both adhesive and anti-adhesive functions. It is expressed on most leukocytes except resting B lymphocytes. Upon activation, proteolytic processing decreases surface expression. CD43 is involved in the activation of T cells, B cells, NK cells, and monocytes. Its counter-receptor, CD169/SIGLEC-1, is expressed on macrophages.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.4 µg) per test (100 µL final volume). Cell number: 10^5–10^8 cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10^3–10^8 cells).

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It contributes to a repulsive barrier that interferes with cellular adhesion but can also promote leukocyte aggregation. Through interactions with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte trafficking. It modulates LFA-1 adhesiveness and signaling. CD43 expression can induce tumor suppressor p53, but in p53/ARF-deficient cells, it may promote tumor proliferation. CD43 dysfunction is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
