Description:
Monoclonal antibody eBio84-3C1 reacts with CD43 (leukosialin, galactoglycoprotein, sialophorin). CD43 is a sialomucin with both adhesive and anti-adhesive properties, expressed on most leukocytes except resting B lymphocytes. Upon activation, proteolytic processing decreases surface expression. CD43 is involved in activation of T cells, B cells, NK cells, and monocytes. Its counter-receptor is CD169/SIGLEC-1, expressed on macrophages.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It forms a repulsive barrier that interferes with adhesion but can promote leukocyte aggregation in some contexts. Through interactions with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and lymphocyte trafficking. It modulates LFA-1 adhesiveness and affects p53-related tumor suppression pathways. Dysregulation of CD43 is linked to Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
