Product Specific Information

The monoclonal antibody 10F7MN recognizes human glycophorin A (CD235a), detecting both M and N alleles. Glycophorin A is a 151 amino acid sialoglycoprotein present on erythrocyte and erythroid progenitor membranes (~500,000 copies per cell). The gene resides on chromosome 4 with two allelic forms (M and N) differing by two amino acids (M: Ser1, Gly5 / N: Leu1, Glu5). Variations may result from toxin exposure and are associated with disease predispositions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells.
Use 5 µL (0.8 µg) per test in a final volume of 100 µL. Cell count range: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Blocking Buffer Recommendation:
Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before mixing with cells. For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD235a (Glycophorin A, GPA) is a single-pass membrane sialoglycoprotein expressed on mature erythrocytes and erythroid precursors. It carries M and N blood group specificities and provides a mucin-like barrier preventing red blood cell aggregation. CD235a also functions as a receptor for Sandei virus, parvovirus, and Streptococcus adhesin Hsa.
