Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43 (leukosialin, galactoglycoprotein, sialophorin), a sialomucin with both adhesive and anti-adhesive functions. CD43 is expressed on most leukocytes except resting B lymphocytes. Upon activation, proteolytic processing decreases surface expression. CD43 participates in activation of T cells, B cells, NK cells, and monocytes. Its counter-receptor is CD169/SIGLEC-1, expressed on macrophages.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg) per test in 100 µL final volume. Cell number: 10^5–10^8 cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample prior to combining with cells.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It regulates cell adhesion and morphology through interactions with actin-binding proteins such as ezrin and moesin. CD43 signaling modulates LFA-1 adhesiveness and influences tumor suppressor p53 activity. Dysregulation is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
