Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43 (leukosialin, galactoglycoprotein, sialophorin). CD43 is a sialomucin with adhesive and anti-adhesive functions, expressed on most leukocytes except resting B lymphocytes. Activation leads to proteolytic reduction of surface expression. CD43 participates in activation of T cells, B cells, NK cells, and monocytes. Its counter-receptor is CD169/SIGLEC-1 on macrophages.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.2 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.

NovaFluor conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, employing nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with high negative charge, expressed on most hematopoietic cells. It modulates leukocyte adhesion and aggregation, interacts with actin-binding proteins (ezrin, moesin), and regulates T-cell morphology and trafficking. CD43 signaling influences LFA-1 adhesiveness and can both enhance and counteract other receptor-induced effects. It is linked to p53 activation and tumor cell regulation. CD43 dysfunction is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
