Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

CA125 determinant is present on a high molecular weight, mucin-like glycoprotein. It is expressed in:

Amnion and derivatives of coelomic and mullerian epithelium (pleura, pericardium, peritoneum)
Adult tissues: epithelial cells of fallopian tube, endometrium, endocervix, pancreas, colon, gall bladder, stomach, kidney, apocrine sweat gland, and mammary gland
Mesothelial cell lining of pleura, pericardium, and peritoneum
Ovarian tumors (serous, endometrioid, clear cell types) and adenocarcinomas of mullerian type
