Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

CA125 determinant is present on a high molecular weight, mucin-like glycoprotein. It is found in:

Amnion and derivatives of coelomic and mullerian epithelium (pleura, pericardium, peritoneum)
Epithelial cells of fallopian tube, endometrium, endocervix, pancreas, colon, gall bladder, stomach, kidney, apocrine sweat gland, and mammary gland
Mesothelial cell linings of pleura, pericardium, and peritoneum
Ovarian tumors of serous, endometrioid, or clear cell types, and adenocarcinomas of mullerian type
