Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also acts as a receptor for thrombospondin and influences T cell activation or apoptosis in the presence of thrombospondin.

This antibody has neutralizing activity and is supplied with 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometry, immunoprecipitation, western blotting, and immunohistochemistry (FFPE sections).
Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (1 µg) per test in 100 µL total volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD47 (Integrin-Associated Protein, IAP) is a transmembrane glycoprotein that interacts with integrins and regulates signaling pathways, including G protein activation and calcium signaling during cell adhesion.
It serves as a receptor for thrombospondin and modulates membrane transport and signal transduction.
CD47 expression is broad across tissues and acts as a ligand for CD172a (SIRPα) on macrophages, preventing phagocytosis of CD47-positive cells.
It also influences cell migration, B/T cell activation, chondrocyte response to mechanical signals, and neuronal development, being abundant in synapse-rich brain and retinal regions.
