Product Specific Information

The monoclonal antibody B6H12 reacts with CD47 (integrin-associated protein, IAP, neurophilin). CD47 is a glycosylated five-transmembrane protein involved in adhesion via interaction with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin and influences T cell activation or apoptosis.

This antibody exhibits neutralizing activity and is supplied with CellBlox Plus Blocking Buffer for optimal staining with NovaFluor conjugates.

Applications Tested

• Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
• Recommended use: 5 µL (1.0 µg) per test (100 µL total volume).
• Cell number: 10⁵–10⁸ cells/test.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample.

Target Information

CD47 (Integrin-associated protein, IAP) is a transmembrane glycoprotein involved in cell adhesion and signal regulation through integrin interactions. It modulates G protein signaling, calcium flux, and serves as a receptor for thrombospondin. CD47 interacts with CD172a (SIRPα) on macrophages to inhibit phagocytosis of CD47-positive cells. It also influences cell migration, T/B cell activation, chondrocyte responses, and neuronal development.
