Product Specific Information

The monoclonal antibody B6H12 reacts with CD47 (also known as integrin-associated protein, IAP, or neurophilin).
CD47 is a glycosylated five-transmembrane protein involved in adhesion via SIRP interaction and associated with β3 integrins (CD51/CD61, CD41/CD61). It mediates bidirectional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also acts as a receptor for thrombospondin and is expressed on hematopoietic and non-hematopoietic cells.

This antibody has reported neutralizing activity.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis and IHC staining of FFPE tissues.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells at 5 µL (1.0 µg)/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.

• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology, employing nucleic acid dye structures engineered for optimized spectral performance and minimal spillover.

Target Information

CD47 (integrin-associated protein, IAP) is a cell surface transmembrane glycoprotein that interacts with integrins and modulates signaling pathways, including G-protein activation and calcium signaling upon adhesion. It serves as a receptor for thrombospondin and regulates membrane transport and signal transduction.
CD47 binds to CD172a (SIRPα) on macrophages, inhibiting phagocytosis of CD47-positive cells. It is involved in cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, with high expression in synapse-rich brain and retina regions.
