Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP (signal regulator protein) and associated with β3 integrins (CD51/CD61 and CD41/CD61). This interaction mediates bi-directional signaling to modify neural synaptic activity and regulate macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin.

T cell expression of CD47 can mediate activation or apoptosis depending on thrombospondin levels. Stimulation by monoclonal antibody has been shown to induce CD4⁺CD25⁻ suppressive activity and increase Foxp3 expression. CD47 is expressed in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and also in non-hematopoietic cells.

This antibody has reported neutralizing activity.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Tested

Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (1.0 µg) per test (in 100 µL final volume).
Applicable cell range: 10⁵–10⁸ cells/test.

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample (regardless of number of NovaFluors).
    • For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein interacting with several integrins and modulating their functions. It regulates intracellular calcium concentration upon cell adhesion and acts as a receptor for the C-terminal domain of thrombospondin.

CD47 serves as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages that prevents phagocytosis of CD47-positive cells. The CD47–CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, particularly in synapse-rich brain and retinal regions.
