Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bidirectional signaling affecting neural synaptic activity and macrophage phagocytosis, and serves as a receptor for thrombospondin.

  • Expression found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic tissues.
  • Reported to have neutralizing activity.
  • Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Usage Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).
    • For antibody cocktails, add CellBlox Plus before combining with cells.

Target Information

CD47 (Integrin-associated protein, IAP) is a transmembrane glycoprotein that interacts with integrins and regulates signal transduction.
It modulates G-protein activation, calcium influx during adhesion, and serves as a receptor for thrombospondin.
CD47 binds CD172a (SIRPα) on macrophages, preventing phagocytosis of CD47-positive cells.
It influences cell migration, B cell adhesion, T cell activation, chondrocyte response to mechanical stimuli, and neuronal development, particularly in synapse-rich regions of the brain and retina.
