Tabalumab neutralizes soluble and membrane-bound BAFF, inhibiting BAFF binding to BR3, TACI, and BCMA. Eight transgenic mice producing serum antibody titers to human BAFF were injected intravenously with 10 µg human BAFF in phosphate buffered saline. The spleens were harvested 3 days later and fused with myeloma cells. Twenty hybridomas were tested for specific binding to human BAFF and human immunoglobulin expression via ELISA. The hybridoma producing the antibody that became tabalumab was originally a human IgG1 isotype, later engineered to a human IgG4 isotype for complete BAFF neutralization without depletion of BAFF-expressing cells.
