Immunogen

Murine antibodies were produced by immunising C3H/HeJ mice with swine peripheral blood lymphocytes. Hybridomas were generated by obtaining splenocytes from the immunised mice and fusing these with the non-Ig-producing myeloma cell line P3X63Ag8.653 via established methods.

Product Specific Information

Hybridoma 898H2-6-15 was shown via double-colour flow cytometry analysis to bind swine lymphocyte populations specifically. FACS analysis confirmed that purified 898H2-6-15 binds an epitope on CD3e shared by another anti-pig CD3 antibody (STH164).
CD3e is crucial for T cell receptor-dependent signal transduction across the lymphocyte membrane.

Target Information

The CD3 complex is essential for transducing antigen-recognition signals into T cell cytoplasm and regulating TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on chromosome 11.
CD3 plays roles in TCR-induced growth arrest, cell survival, and proliferation. It is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
