PA1-757 detects transfected beta-secretase 1.
It has been successfully used in ELISA, ICC/IF, Western blot, and immunoprecipitation applications.
The molecular weight of BACE1 detected may vary depending on the lysate tested, which may be due to glycosylation differences among cell lines.
The PA1-757 immunizing peptide (Cat. # PEP-093) is available for neutralization and control experiments.

Target Information

BACE1 (beta-secretase) is a key enzyme involved in the production of amyloid beta-peptides (Abeta) found in extracellular amyloid plaques of Alzheimer’s disease (AD).
In some cases, early-onset familial AD can be attributed to a “Swedish” mutation in the amyloid precursor protein (APP), which enhances cleavage by BACE1.
Therapeutic approaches have focused on inhibiting BACE1 and other APP-cleaving enzymes, such as gamma-secretase.
APP is first cleaved by beta-secretase, producing a soluble derivative and a membrane-anchored 99-amino acid fragment (C99).
C99 serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide, which accumulates in brains of Alzheimer’s patients.
BACE1 (Asp2) is a transmembrane aspartic protease that colocalizes with APP.
