Product Specific Information
PA1-760 detects serotonin N-acetyltransferase (AA-NAT) from rat tissues.
It has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~23 kDa protein representing unphosphorylated and phosphorylated AA-NAT from rat pineal gland whole protein extract.
PA1-760 immunizing phosphopeptide (Cat. # PEP-064) corresponds to amino-terminal amino acid residues 24-33 of mouse AA-NAT.
Total anti-amino-terminal AA-NAT-specific IgG (Cat. # PA1-760) was affinity purified on resin-coupled non-phosphopeptide (Cat. # PEP-063).
Phospho-specific amino-terminal IgG (Cat. # PA1-761) was subsequently isolated on resin-coupled phosphopeptide (Cat. # PEP-064).
This sequence is completely conserved between mouse, rat, and human AA-NAT protein.
Target Information
Arylalkylamine N-Acetyltransferase (AANAT) is the penultimate enzyme in melatonin synthesis.
Hormonal melatonin has been implicated in sleep and circadian clock function.
Large changes in the abundance of AANAT are believed to be responsible for the day/night rhythms in melatonin synthesis in the pineal gland and retina.
Most research shows that AANAT activity is regulated by controlling the steady-state levels of the protein.
Recent studies suggest that melatonin production can also be controlled by mechanisms independent of AANAT protein abundance, possibly explaining the rapid physiological increase in circulating melatonin at the onset of darkness in humans.
