Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas.

The extracellular region contains seven Ig-like domains capable of binding sialic acid–containing ligands, while the intracellular region includes six tyrosine residues within ITIM and activation-like motifs. These motifs are phosphorylated upon B-cell receptor engagement, regulating both positive and negative B-cell receptor signaling.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.02 µg) per test in 100 µL final volume, with cell counts between 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL of CellBlox Plus per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures that enable engineered fluorescence with minimized spillover and spectral spread.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed on B-lymphocytes. It appears early in B-cell differentiation, similar to CD19, but its membrane expression is limited to mature B cells (CD22⁺) and absent in plasma cells (CD22⁻).
CD22 is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte-predominant Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.

It preferentially binds α2,6-linked sialic acid. Ligand-induced tyrosine phosphorylation of CD22 is involved in B-cell receptor signaling regulation, acting both as a positive regulator (via Src family tyrosine kinases) and as an inhibitory receptor (via recruitment of cytoplasmic phosphatases).
