Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily.
CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (130 and 140 kDa) produced by alternative splicing.
It is highly expressed on mature B cells and B cell lymphomas. The extracellular region contains seven Ig-like domains capable of binding sialic acid moieties on epithelial, endothelial, B and T cells.
The intracellular region contains six tyrosine residues within ITIM and activation-like motifs that regulate B-cell receptor signaling.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.2 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

Each NovaFluor conjugate or kit ships with CellBlox Blocking Buffer.
Use 5 µL per sample when staining with NovaFluor conjugates, including single-color compensation controls.
For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures and reduced spillover.

Target Information

CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in both cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation and is useful for phenotyping mature leukemias.
Strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds α2,6-linked sialic acid and participates in B-cell receptor signaling regulation via tyrosine phosphorylation.
