Product Specific Information

Description:
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two chains, CD22α and CD22β (130 and 140 kDa), produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas. The extracellular region contains seven Ig-like domains capable of binding sialic acid ligands on epithelial, endothelial, B, and T cells. The intracellular region includes tyrosine residues within ITIM and activation-like motifs that regulate B-cell receptor signaling.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures, designed to minimize spillover and spread in multicolor flow cytometry.

Target Information

CD22 (BL-CAM) is a 130–140 kDa type I membrane glycoprotein expressed in B-lymphocytes. It appears during B-cell differentiation alongside CD19 and is limited to mature B cells (CD22+) and absent in plasma cells (CD22−). CD22 is strongly expressed in hairy cell leukemia and can aid in phenotyping mature leukemias. It binds preferentially to α2,6-linked sialic acid and plays a role in regulating B-cell receptor signaling by interacting with tyrosine kinases and phosphatases. CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
