Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7, also known as gp40 and Leu9. CD7 is a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chain sequences, while the proline-rich C-terminal region forms a stalk projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a small subset of normal B cells, and malignant B cells. Cross-linking of CD7 modulates T and NK cell activity, affecting calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 interacts with phosphoinositol 3'-kinase, leading to D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high levels of CD7 can generate lymphoid cells (B, NK, dendritic) but not myeloid or erythroid cells, distinguishing primitive lymphoid progenitors from pluripotent stem cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Normal human peripheral blood cells at ≤5 µL (0.8 µg)/test (100 µL final volume, 10^5–10^8 cells/test). Optimize antibody titration for best results.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10^3–10^8 cells).

NovaFluor conjugates utilize Phiton™ technology, offering engineered fluorescent signatures optimized for minimal spillover and spread.
