Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chains, while the C-terminal region is proline-rich and forms a stalk projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and a minor population of normal and malignant B cells. Cross-linking CD7 modulates T and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3'-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels can generate lymphoid lineages (B-cells, NK cells, dendritic cells) but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive human lymphoid progenitors from pluripotent stem cells.

Applications Tested

Tested by flow cytometry on normal human peripheral blood cells at ≤5 µL (0.8 µg)/test in 100 µL final volume. Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before mixing with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimal spectral performance and minimal spillover.

Target Information

CD7 (gp40, Leu9) is a 40 kDa immunoglobulin superfamily member expressed on most T lymphocytes and T cell leukemias. It is also found on NK cells and subsets of B cells. CD7 cross-linking modulates immune cell activity and is associated with phosphoinositol 3'-kinase signaling. CD7 expression is an important marker in leukemia diagnostics.
