Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin gene superfamily.

  • The N-terminal region (aa 1–107) is homologous to Ig kappa light chain, while the C-terminal region is proline-rich.
  • CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and a small subset of normal and malignant B cells.
  • Cross-linking of CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation.
  • CD7 associates with phosphoinositol 3'-kinase and induces D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of CD34(+) CD38(-) cord blood cells expressing high CD7 levels has been identified, capable of generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. The CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.

Important Notes

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) with NovaFluor conjugates for optimal staining.
    • Add 5 µL per sample, regardless of panel complexity.
    • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor Dye Technology

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures designed for engineered fluorescent signatures with minimized spillover and spread.
