Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and also in various normal and malignant cells. CD146 functions as a calcium-independent cell adhesion molecule and may act in signal transduction by recruiting Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly. It plays a role in maintaining endothelial cohesion and may facilitate melanoma cell interaction with vascular elements, enhancing tumor spread. CD146 is also active in neural crest cells during embryonic development and acts as a surface receptor triggering tyrosine phosphorylation of FYN and PTK2, with transient intracellular calcium increase.
