Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell–cell junctions in endothelial cells and in various normal and malignant cell types. CD146 functions as a calcium-independent cell adhesion molecule and may also act in signal transduction through Fyn kinase activation and tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly. It contributes to endothelial cohesion and may facilitate melanoma cell interaction with vascular elements, promoting tumor spread. CD146 may also play a role in neural crest cell adhesion during embryonic development and acts as a surface receptor triggering phosphorylation of FYN and PTK2, leading to transient increases in intracellular calcium concentration.
