Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in all endothelial cells and in various normal and malignant cell types. In addition to functioning as a calcium-independent cell adhesion molecule, CD146 may act in signal transduction by recruiting Fyn kinase and promoting tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
CD146 contributes to endothelial cell cohesion and may facilitate melanoma cell interaction with vascular elements, enhancing tumor spread. It also functions as an adhesion molecule in neural crest cells during development and as a surface receptor triggering tyrosine phosphorylation of FYN and PTK2, leading to transient increases in intracellular calcium concentration.
