Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal and malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may also act as a signal transduction molecule, recruiting Fyn kinase and triggering tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a role in endothelial cohesion and melanoma cell interaction with vascular elements, enhancing tumor spread. CD146 may also be active in neural crest cells during embryonic development and acts as a surface receptor inducing tyrosine phosphorylation of FYN and PTK2 and transient intracellular calcium increase.
