Description:
The monoclonal antibody 3D12 (HLA-E) recognizes human HLA-E, a member of the Human Leukocyte Antigen family. HLA-E is an MHC Class IB molecule involved in NK cell recognition. It binds a restricted set of peptides derived from other Class I leader sequences, forming a complex with β2-microglobulin (B2M) that interacts with NKG2A on NK cells, protecting HLA-G-expressing cells from NK cell-mediated killing. HLA-E expression is ubiquitous.

Applications Tested:
This antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is ≤1 µg per test (defined as the amount required to stain cells in 100 µL final volume). Optimal titration is advised for specific assays.

Target Information

HLA-E belongs to the HLA class I heavy chain paralogues. It forms a heterodimer with β2-microglobulin, with the heavy chain anchored in the membrane. HLA-E binds peptides derived from other class I molecule leader sequences.
The heavy chain (~45 kDa) gene contains 8 exons encoding distinct structural domains:

  • Exon 1: Leader peptide
  • Exons 2–3: α1 and α2 domains (peptide binding)
  • Exon 4: α3 domain
  • Exon 5: Transmembrane region
  • Exons 6–7: Cytoplasmic tail

HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens via HPLC and tandem mass spectrometry.
