Description:
The ISA-3 monoclonal antibody reacts with human ICOS (Inducible COStimulatory molecule), also known as H4, CRP-1, and AILIM. ICOS is a T cell-specific activation molecule and a third member of the CD28/CTLA-4 family. Human ICOS (55–60 kDa) consists of 27 kDa and 29 kDa chains and is essential for T cell activation and humoral immune responses, particularly in memory B cell and plasma cell generation.

ICOS binds to its ligand B7h/B7RP-1 on activated APCs and inflamed tissues. Plate-bound ISA-3 provides costimulation for T cells, inducing cytokines such as IL-4, IL-5, and IL-10 but not IL-2. ISA-3 shares reactivity with clone F44.

Clone C398.4A (anti-mouse ICOS/H4, Product #14-9949-82) cross-reacts with human ICOS but binds a different epitope. For the brightest staining of activated human T cells, use Product #13-9948-82 or #12-9948-42 rather than #11-9948-42.

• Immunoprecipitation
• Immunohistochemical staining of frozen tissue sections
• In vitro functional assays (use Functional Grade purified ISA-3, Product #16-9948-82)

Applications Tested:
Tested by flow cytometric analysis of unstimulated and PHA-activated human blood cells. Recommended ≤0.5 µg per test in 100 µL volume. Cell number: 10⁵–10⁸ cells/test. Optimize dilution for specific assays.
