Product Specific Information

The monoclonal antibody 87G recognizes human HLA-G, a nonclassical MHC type involved in inhibiting immune responses.
HLA-G has seven isoforms, and antibody 87G recognizes both HLA-G1 and soluble HLA-G5.

Expression of HLA-G is found primarily in fetal trophoblast cells, protecting the fetus from maternal immune attack.
Abundant HLA-G expression is also identified in certain tumors such as melanoma, breast carcinoma, renal carcinoma, and hematologic malignancies (CLL, AML, B-CLL).
Additional expression is seen in pancreatic islets, erythroid and endothelial progenitors, and adult thymic medulla.

HLA-G⁺ CD4 or CD8 cells in normal peripheral blood act as regulatory cells with a unique cytokine profile.
Receptors for HLA-G include CD85j/ILT2, CD85d/ILT4, and CD158.
Recent studies show a role for HLA-G in transplant tolerance and maintenance.

Target Information

HLA-G belongs to the HLA class I heavy chain paralogues.
It forms a heterodimer of a heavy chain and beta-2 microglobulin.
The heavy chain (~45 kDa) gene contains 8 exons:

  • Exon 1: Leader peptide
  • Exons 2–3: α1 and α2 domains (peptide binding)
  • Exon 4: α3 domain
  • Exon 5: Transmembrane region
  • Exon 6: Cytoplasmic tail

HLA-G is expressed on fetal-derived placental cells and plays a key role in immune tolerance.
HLA/MHC antibodies are crucial for Immunopeptidomics, enabling identification and characterization of neoantigens via LC-MS/MS.
