Product Specific Information

This GzA-3G8.5 monoclonal antibody reacts with mouse Granzyme A, the most abundantly expressed of the ten granzyme serine proteases in mice. Granzymes are proteins released from NK cells and cytotoxic T lymphocytes that induce death in target cells by cleavage of intracellular substrates, playing a critical role in immune defense against viruses, tumors, and intracellular bacteria.

Granzyme A activates caspase-independent cell death pathways morphologically similar to apoptosis, characterized by mitochondrial and DNA damage. It may also contribute to inflammation by processing pro-IL-1β. Granzyme A shares overlapping substrate specificity with Granzyme K, explaining the minimal cytotoxicity reduction in Granzyme A-deficient CTLs.

Applications Tested:

  • Tested by intracellular staining and flow cytometric analysis of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol.
  • Recommended ≤ 0.06 µg per test (100 µL final volume).
  • Cell number: 10⁵–10⁸ cells/test (optimize empirically).
  • Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, or Yellow-Green.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells recognize, bind, and lyse specific target cells bearing nonself antigens, typically peptides or proteins from intracellular pathogens. Granzyme A is a T cell- and NK cell-specific serine protease functioning as a key component in target cell lysis.
