Target Information

GLTSCR2 (also known as NOP53) is a nucleolar protein involved in the integration of the 5S RNP into the ribosomal large subunit during ribosome biogenesis. It may also play a role in rRNA transcription and act as a nucleolar sensor regulating p53/TP53 activation in response to ribosome biogenesis perturbation, DNA damage, and other stress conditions.

Disruption of the interaction between NOP53 and RPL11 allows RPL11 transport to the nucleoplasm, inhibiting MDM2 and enabling p53/TP53 activation. GLTSCR2 may positively regulate p53/TP53 in cell cycle arrest and apoptosis by preventing its MDM2-dependent degradation. Initially identified as a tumor suppressor, it can influence cell proliferation and apoptosis by stabilizing PTEN and antagonizing the PI3K-AKT/PKB signaling pathway. It may also interact with NF2 to inhibit proliferation and increase apoptosis, and regulate NPM1 localization and degradation, thereby affecting MYC-NPM1–mediated transcription. Additionally, it may regulate cellular aerobic respiration and modulate interferon-beta response during viral infection by inhibiting DDX58/RIG-1.
