Product Specific Information

Description:
Monoclonal antibody eBio3A3-N2 reacts with human IL-21, a potent stimulator of NK and cytolytic T cells. IL-21 (131 amino acids) is closely related to IL-2 and IL-15 and is expressed in activated CD4+ cells. It stimulates B-cell proliferation via CD40 antigen crosslinking, bone marrow progenitor proliferation, and NK-cell marker CD56 expression in presence of IL-15.

Applications Reported:
Validated for intracellular staining and flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining of restimulated, Th17-polarized human peripheral blood cells. Recommended use: 5 µL (0.125 µg)/test in 100 µL final volume. Cell number may vary (10⁵–10⁸ cells/test).

Notes:

  • IL-21 staining shows donor variability; stimulation times should be optimized.
  • Enhanced staining observed in Th17-polarized cells.
  • eFluor® 660 replaces Alexa Fluor® 647 (Emission 659 nm, Excitation 633 nm).
  • Ensure instrument compatibility with red laser detection.
  • Filtration: 0.2 µm post-manufacturing.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine mainly produced by NKT, Th17, and TFH cells. In TFH cells, IL-21 signals through IL-21R and STAT3, activating Bcl6. It is critical for TFH development and function, promoting B-cell differentiation into plasma and memory cells, stimulating IgG production, and inducing apoptosis in naive B cells.

In Th17 cells, IL-21 upregulates IL-23R through STAT3, IRF4, and RORγt, facilitating maturation via IL-23. It also downregulates Foxp3. IL-21 deficiency protects mice from chemically induced colitis, while elevated IL-21 levels are observed in colitis models.
