Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins CD51/CD61 and CD41/CD61. It mediates signaling related to synaptic activity and macrophage phagocytosis and serves as a receptor for thrombospondin.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Use in flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.6 µg) per test (100 µL final volume). Cell number per test: 10⁵–10⁸ cells.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures engineered for optimal fluorescent signatures with reduced spillover and spread.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and regulates their functions. It modulates signaling pathways, including G protein activation and calcium flux during cell adhesion. CD47 also serves as a receptor for thrombospondin and plays roles in signal transduction, immune regulation (via CD172a/SIRPα interaction), cell migration, and neuronal development. Expression is widespread, including hematopoietic and non-hematopoietic cells.
